Research Abstract

1.DNA固定化磁性ビーズの作製とホール素子による検出
単鎖DNAをGMA被覆フェライトビーズの表面に固定化ことに成功した。これと相補的な単鎖DNAをホール素子上に固定し、両者のハイブリッダイゼーションによって、DNAの定量的検出が可能であることを示した。
2.新規MRI造影剤の開発
高い造影効果を持つフェライト造影剤を開発するために、フェライト表面を多彩に修飾・加工する技術を開発した。その結果、フェライトを芯に持ち、粒径が約20nmで、表面が特定組換えタンパク質で被覆された新たな造影剤候補物質を開発した。
4.抗がんハイパーサーミア効果の検証
粒径が17mmのフェライト粒子を用いて、ヒトの大腸癌細胞colo205を、非侵性の周波数180Hzの交流磁界によって誘導加熱し、その殺傷効果を確認し、非侵性抗がんハイパーサーミアへの応用の可能性があることを明らかにした。
5.タンパク質固定化磁性ビーズの作製とバイオスクリーニング
タグ付きプロテインGを含む組換えタンパク質を、タグを介して磁性ビーズに固定化する方法を確立し、リガンドの標的となる組換えタンパク質を直接、あるいは抗体であれば何でも、プロテインGを介して間接的に磁性ビーズ表面に捕捉してスクリーニングできる系を確立した。
6.磁化の強いGMA被覆GMAビーズの開発
フェライトより2倍以上も強い磁化を持つ金属Fe粒子(粒径7-20nm)をSt-GMAおよびGMAで被覆したバイオスクリーニングに適したビーズの作製法を確立した。
