Research Abstract |
The study was carried out for treating the brain of neurodegenerative disorders by transplanting cells to deliver the deficit materials. At first, we established ES cell-derived glial precursor cells. ES cells were grown to be embryoid bodies when cultured with insulin, transferrin, and fibronectin, in the absence of LIF. The cells were spread by trypsinization and continued the culture with insulin, transferrin, progesterone, putrescine, and laminin. The cells became bipolar cells when basic fibroblast growth factor (FGF2) and epidermal growth factor (EGF) were added. The cells were stained immunochemically with anti-A2B5, anti-GFAP, and anti-04 antibodies, and confirmed the cell-differentiations. The ES cell-derived glial precursor cells were transplanted into the brain ventricle of β-galactosidase knock-out mouse at 2 weeks of age. One week after the transplantation, the mice were sacrificed and the brain specimens were stained with X-Gal. However, no X-Gal positive cells were found in the brain. When the cells were transplanted in the ventricle of newborn mice, a few X-Gal positive cells were clotted in the brain at one week after the plantation.
