Research Abstract |
Forty-one gastric cancer specimens (20 were differentiated adenocarinoma and 21 undifferentiated) were analyzed in this study. Using Manual tissue arrayer, 2 grafts (2mm in diameter), which showed representative histological feature, were taken out as from each specimen ("donor block"). Forty-eight grafts were put per "recipient block". The graft was lined up in the donor block by histological type and tumor stage. In cutting the recipient block, successfully cut into 3 micro-meter thickness without the grafts in the recipient block being destroyed. It became clear that an array block well reflected the histological structure of donor blocks in tumor cell and gland density when it was compared with donor blocks. In immunohistochemistory, a positive correlation was observed in p-53, Ki-57 and COX-2 expression intensity between donor and microarray (p<0.0005, <0.0005 and <0.0001, respectively). Therefore, diagnostic system by immunohistochemistory using Tissue Microarray was supposed to be useful for human gastric cancer We are now evaluating the efficacy of Tissue Microarray by analyzing the expression of other proteins, such as VGEF-C, thymidylate synthase, dihydropyrimidine dehydrogenase, thymidine phosphorylase and so on..
