Research Abstract |
Selection of appropriate protocols for treatment of superficial esophageal squamous cell carcinoma (SESCC) is dependent on lymph node metastasis status. Previously, we examined the relation between DNA sequence copy number aberrations detected by comparative genomic hybridization (CGH) and lymph node metastasis in 31 surgically resected SESCCs. In the result, the gains of 8q24 and/or 20q12-qter appears to be associated with the gain of lymph node metastasis in patients with SESCC. We hypothesis that there are some important oncogenes or antioncogenes about progression of squamous cell carcinoma of esophagus was strong in these chromosome region. about lymph node metastasis, used BAC(Bacterial Artificial Chromosome) library and made DNA microarrays of superscription chromosomal aberration area In this study, used BAC (Bacterial Artificial Chromosome) library we made DNA microarrays of 8q24 and 20q12-qter chromosomal aberration area and examined an unknown cancer-related genes. We did chromosome CGH analysis for four cases with lymph node metastasis, five cases without lymph node metastasis in SESCC newly. Furthermore, it was high-frequency in two chromosome region that we stated above by array CGH analysis using the genome DNA array that analysis in a gene level was possible and tried the genetic search that aberration occurred in. By chromosome CGH analysis of nine cases that we did newly this time, high-frequency amplification was confirmed in 2p, 3q, 5p, 7p, 8q20, 20q12-qter same as conventional analysis results. Although 8q24, 20q12-qter was high-frequency and amplified it, the correlation was not found in this investigation between these area and lymph node metastasis of superficial squamous cell carcinoma of esophagus. We were able to identify 45 SESCC-related candidate gene cluster when we analyzed 8q24, 20q12-qter area in 21 SESCC cases in detail. In addition, 26 unknown genes were included in these.
