舌発がんにおける炎症関連酵素、PPARsの関与とその制御による発がん防御

Research Abstract

炎症と舌発がんの係わりを明らかにする目的で動物実験を実施した。
雄性F344ラット(4週齢)20匹に4-nitroquinoline 1-oxide(4-NQO)を20ppmの濃度で8週間飲水投与し、実験開始後32週で犠牲死させた。舌を摘出し、その病理学的解析を行った。同時に、非腫瘍部および過形成病変、異形成病変、扁平上皮がんを含む舌組織におけるIL-α、ILβを測定し、各組織中の肥満細胞と新生微小血管の数を測定した。
その結果、IL-α(pg/mg)は、非腫瘍部:7.8±1.5、過形成病変:13.8±4.6、異形成病変:37.8±7.9、扁平上皮がん:45.1±7.7、ILβ(pg/mg)は非腫瘍部:0.81±0.18、過形成病変:1.43±0.48、異形成病変:3.92±0.59、扁平上皮がん:4.62±0.61、tryptase陽性肥満細胞数:(/mm^2)は、非腫瘍部:6.5±1.6、過形成病変:9.9±2.0、異形成病変:18.4±2.0、扁平上皮がん:28.4±5.2、CD31陽性新生微小血管数(/mm^2)は、非腫瘍部:18.0±1.8、過形成病変:17.8±4.6、異形成病変:28.4±5.8、扁平上皮がん:51.8±7.2といずれの値も発がん過程の進展に伴って有意(P<0.05)な増加をみた。
これらの結果から、舌発がんの進展、特に悪性転化に炎症が関わることが判明した。また、肥満細胞数の増加と血管新生の相関が示唆され、特に癌細胞の浸潤部では、肥満細胞の増加が観察された。このように、過形成、異形成、癌の侵入部最前線では、炎症性肥満細胞は、間質を再編成し、脈管形成を促進するために動員されることが示唆された。
