Research Abstract

本研究ではRAB27A遺伝子の欠損・変異により引き起こされるヒト遺伝病Griscelli syndromeの発症メカニズムを分子レベルで解明することを目的としている。本年度はGriscelli syndrome患者に特徴的な毛髪の部分白化(メラノソーム輸送異常)に焦点を当て、メラノソーム輸送の分子メカニズムを解明することに成功した(J.Biol.Chem.(2003)278,11386-11392;Mol.Cell.Biol.(2003)23,5245-5255;J.Cell Sci(2004)117,583-591)。すなわちメラニン色素産生細胞においてはRab27AエフェクターSlac2-aが豊富に発現しており、Slac2-aはアミノ末端側のSHD領域を介してメラノソーム上のRab27Aと、またカルボキシル末端側を介してモーター蛋白質であるミオシンVa及びアクチン線維と四者の複合体を形成し、メラノソームを核周辺から細胞膜周辺へと輸送することが明らかとなった。また、部位特異的アミノ酸置換によりこれらのリガンドを結合出来ない(四者複合体を形成しない)Slac2-aの変異体をメラニン色素産生細胞に発現させると、dominant negative効果によりGriscelli syndrome患者と同じ症状を細胞レベルで誘発することにも成功した。さらに、内分泌細胞においてはSlac2-aとは異なるRab27Aエフェクターが発現し(Slac2-c,Slp4-a)、ホルモン分泌を制御することも明らかになった(J.Biol.Chem.(2003)278,15373-15380;J.Biol.Chem.(2003)278,15390-15396;Mol.Biol.Cell(2003)14,4103-4113)。
