Outline of Annual Research Achievements

Experiment 1: We investigated the effects of Manuka Honey (MH) by dividing 6-week-old male C57BL6/J mice into three groups (n=5) with equal mean body weights: (1) AIN-93G basal diet and normal drinking water for the course of the entire experiment (CON group), (2) 10% MH- supplemented AIN-93G basal diet and normal drinking water for the course of the entire experiment (MH group) (3) AIN-93G basal diet and normal drinking water for 1 week, after which colitis was induced by administering 2% (w/v) dextran-sodium-sulphate (DSS);containing drinking water (DSS group); and (4) 10% MH (Comvita Inc., Auckland, New Zealand)-supplemented AIN-93G basal diet and normal drinking water for 1 week, then 2% DSS-containing drinking water (DSSMH group) for 10 days. MH intervention significantly suppressed colon shortening.
Experiment 2: We practiced our methods of colon cancer induction by dividing 6-week-old male C57BL6/J mice into two groups (n=5) with equal mean body weights. The AOMDSS mice were given a single intraperitoneal injection of Azoxymethane (AOM, 10 mg/kg body weight), followed by administering 7 days of tap water and then administering 2% (DSS) in drinking water for 7 days. The tap water and DSS drinking drinking water cycle was repeated thrice. Normal control mice were injected with saline and subjected to tap water throughout the entire experiment. At the end of the experiment, the colon was excised and the number of tumors counted. There was a significantly larger number of tumours in the AOMDSS mice. We had successfully created a model of AOMDSS induced colon cancer.

2: Research has progressed on the whole more than it was originally planned.

Reason

We have completed verifying the anti-colitic effects of MH honey in a DSS-induced colitic mice model. We have also completed creating a AOM-DSS induced colon cancer model in our laboratory.

Strategy for Future Research Activity

We will perform dose-dependent investigation of anti-colitic effects of MH dietary supplementation. In our current experiment, we used 10% MH supplemented AIN-93G diet and we will repeat the experiment using 5% and 10% MH.
We will also investigate the anti-colon carcinogenic effects of 5% and 10% MH supplementation.
