Outline of Annual Research Achievements

子宮内膜症はエストロゲン依存性の炎症性疾患である。女性の10％程度が罹患しており、quality of lifeを著しく損なっている。しかしながらその治療法は確定していない。我々はゲノム全域アソシエーション・スタディの結果から得られたSNPの機能解析を目指し、アレルに伴う遺伝子発現変化を検討している。対象としたのは9p21で得られたシグナルで、エンハンサー活性に影響を与えると予想され、ターゲットとなる遺伝子はnon-coding RNAのANRILである。すでにアレルに伴いANRIL発現が変化することを示すことができている(PLOS Genet 2016)。これらはアレルによりエンハンサー活性の違いが認められることをクロマチン三次元構造などで示したものである。しかしながらin vitro studyであり、in vivoでのエンハンサー活性は不明であった。そこで、エンハンサー活性検討システムをゼブラフィッシュで確立した。このシステムでは、SNPを含むエンハンサー候補領域をGFP遺伝子の上流にクローニングし、これをゼブラフィッシュの受精卵にマイクロインジェクションすることで、in vivoでのエンハンサー活性をGFPシグナルとして検出可能にした。
このように疾患に関連するSNP解析からアレルに伴うエンハンサー活性の違い、そしてin vivoでのエンハンサー活性を示すことができ、ありふれた疾患の疾患遺伝子研究に大きく貢献することとなった。
