Outline of Annual Research Achievements

新世界ザルは食性や色覚の多様性が顕著であるため、進化生態学的な文脈で様々な感覚の相互作用を理解するのに好適といえる。そこで、多様な色覚型と食性の新世界ザル6種を対象に、ターゲットキャプチャーと次世代シークエンシングにより、遺伝子数の膨大な嗅覚受容体(OR)に加え、苦味受容体(TAS2Rs)と旨味甘味受容体(TAS1Rs)遺伝子の全レパートリーを明らかにすることにした。トレードオフ仮説の予測と異なり、新世界ザルで唯一恒常的3色型色覚のホエザルは、他の2色型3色型多型の種に比べ、OR偽遺伝子割合は特に高くはなかった。総OR遺伝子数や偽遺伝子割合に種間で顕著な違いが見られない一方で、機能遺伝子と偽遺伝子のレパートリー構成は種間で大きく異なっていた。TAS2Rsは中立対照に対して塩基多型度が、特に非同義変異で高い傾向にあり、苦味感覚の多様化進化が示唆された。これら6種の新世界ザルのTAS2R16とTAS2R38を培養細胞系で再構成したところ、苦味物質に対する反応性に種間で顕著な違いが見られた。一方、TAS1Rsは機能制約が緩んでいる傾向がみられ、タマリン属では旨味受容体TAS1R1が偽遺伝子化していた。これらの結果は、霊長類の進化において、化学物質受容体遺伝子群の機能・偽遺伝子構成が能動的に変化していることを示唆し、化学物質感覚の重要性を物語っていると考えられる。
一方、旧世界ザルのニホンザルTAS1R2/TAS1R3はヒトが感じられない程度の麦芽糖の甘味もショ糖の甘味と同じくらいに感じることを発見した。自然界では、炭水化物はデンプン等の多糖類として穀物や葉に含まれているので、唾液等に含まれる酵素（アミラーゼ）によって分解された結果、麦芽糖が生じる。ニホンザルはこれを強い甘味として感じることで、採食に役立てている可能性が示唆された。

Reason

多様な色覚と食性の新世界ザル6種からのターゲットキャプチャーと次世代シークエンシングから順調に遺伝子数の膨大なOR、TAS2R、TAS1R遺伝子の全レパートリーを明らかにすることができた。その結果、従来の限られて知見から想定されたことと、これらの遺伝子の進化像が異なることを示すことができた。

Strategy for Future Research Activity

旧世界霊長類の多数種とヒトの集団試料に対して、OR、TAS2R、TAS1R遺伝子の全レパートリーに対して、ターゲットキャプチャーと次世代シークエンシングを適用し、霊長類におけるこれらの感覚遺伝子の進化とヒトにおける多様性を明らかにする。
