Outline of Annual Research Achievements

本研究課題においては、イネの高温・高CO2応答の学術的理解を進め、高温・高CO2環境に適応するイネ品種の作出技術を確立し、農業振興・国際競争力の向上に貢献することを目的としている。新潟大学・刈羽村先端農業バイオ研究センターに設備されている人工光・CO2濃度制御型閉鎖温室(L2.7 x W3.0 x H2.9m)を用いて玄米外観品質に及ぼす強光・高CO2濃度(光量800μmol m-2 s-1・CO2 濃度1600 ppmv)および高温・高CO2濃度(温度L30℃/D25℃・CO2 濃度1600 ppmv)のパルス処理による影響を調べた。なお、コントロール条件は光量450μmol m-2 s-1・温度L26℃/D23℃・CO2 濃度400 ppmvとした。開花から登熟期初期において玄米外観品質に及ぼす強光・高CO2濃度および高温・高CO2濃度の感受性が高いことが分かった。また、高CO2濃度環境下のみでは顕著な玄米白濁化は起こらないが、高CO2濃度は高温ストレスを助長することが明らかになった。さらに、強光・高CO2濃度および高温・高CO2濃度の処理によって生じた白濁部位におけるα-アミラーゼアイソフォームタンパク質の発現を特異抗体を用いたイムノブロッティングによって解析したところ、両処理においてAmyII-4の顕著な増加が確認された。加えて、複合ストレスに適応するイネ品種を開発するため、高温登熟性優良品種「ゆきん子舞」と塩耐性品種「海神」を掛け合わせ、迅速世代促進法により4回の戻し交配を行い、塩耐性を有する「ゆきん子舞」新系統(YNU1)を作出した。高塩濃度環境下でのYNU1の幼芽と幼根のNa+およびK+の動態を電子線マイクロアナライザーを用いて調べたところ、YNU1ではNa+/K+比が低く抑えられていることが分かった。
