| Outline of Annual Research Achievements |
システインパースルフィド (Cys-SSH)に代表される活性イオウ分子種は、価数0のイオウ原子であるsulfane sulfurを有し、高い反応性を持つ。活性イオウ分子種は種々の細胞内タンパク質へ組み込まれ、生理機能を調節していることが報告されている。一方、血清など細胞外のタンパク質に対しては、その存在や機能について殆ど明らかにされていない。これは、細胞外の酸化的環境下における信頼性の高い活性イオウ分子種測定法が確立されていないことが原因である。この課題を克服するため、本研究では、新規活性イオウ分子種測定法の確立ならびに活性イオウ付加アルブミンの構造・機能解析を行った。本年度は、sulfane sulfurの還元剤を構築し、メチレンブルー法と組み合わせた新規の活性イオウ分子種の測定法 (Elimination Method of Sulfide from Polysulfide, EMSP)を開発し、論文化に至った。また、健常血清中の活性イオウ分子種を測定したところ、約8 mMもの活性イオウ分子種が存在していること、また主要な活性イオウ付加タンパク質がヒト血清アルブミン (HSA)であることを同定した。興味深いことに、血清sulfane sulfurレベルは肝硬変疾患などの酸化ストレス疾患で有意に減少し、有病数との間で相関性を示した。以上より、活性イオウ分子種モニタリングは、早期酸化ストレス検出法としての応用が期待される。加えて、HSAを基盤としたsulfane sulfur送達システムは、酸化ストレス疾患の新たな予防・治療戦略の可能性を秘めている。

予定していた実験計画はすべて順調に遂行していることに加え、当初予期していなかった他のタンパク質と活性イオウ種の関係性も明らかにすることができ、当初の計画を越えた進展に繋がっていると考えている。

| Strategy for Future Research Activity |
今年度開発に成功した新規の活性イオウ分子種の測定法 (Elimination Method of Sulfide from Polysulfide, EMSP)法を活用し、様々な疾患との因果関係を明らかにし、病態の早期診断を行うこと、加えて、新たな活性イオウデリバリーシステムを構築し、治療へ応用するという「診断と治療」の両方から研究を推進していく予定としている。診断に関しては、すでに活性イオウ分子種の血清中の正常値を測定していることから、種々の患者血清を用い、網羅的検討を行う。治療に関しては、安定な輸送担体であるヒト血清アルブミンなどを利用し、調製することを予定している。
