Outline of Final Research Achievements |
Cell free DNA (cfDNA) in the blastocoel fluid (BCF) was collected and amplified, and with which genetically analysis was performed. Removal and amplification of cfDNA in BCF was successfully carried out. As a method of whole genome amplification, PicoPlex was suitable for amplification of cfDNA in BCF because it was shown that PicoPlex was superior to MDA in amplification success rate and recovery amount. It seems to be technically difficult to comprehensively and uniformly amplify cfDNA that is slightly present in BCF, so sufficient results in the assessment of numerical abnormality of the all chromosomes using array CGH were not obtained. However, the results demonstrated that diagnosis of monogenic diseases was sufficiently possible.
