Research Abstract |
1.Development of chemical speciation and in situ determination of bioactive trace metals (1)Determination of suspended particulate Co, Ni, Cu, Zn, Cd and Pb has been developed. (2)Determination of dissolved and acid-dissolvable Zr, Hf, Nb, Ta and W has been developed. (3)In-situ flow analysis of alkalinity and total carbonate has been developed. 2.Development of analytical methods for enzymes and proteins of phytoplankton and bacteria (1)Detection of Fe-S protein PsaC and PsbA(D1) in photosynthetic system has been developed. (2)Anti-ferredoxin and anti-flavodoxin antibodies, which cross-react with ferredoxin and flavodoxin of marine diatoms, respectively, have been developed using synthetic peptides in order to estimate their Fe stress. 3.Observation of the variation and relationship of metallome and proteome (1)Different distributions of Fe, Co, Ni, Cu, Zn and Cd in seawater between the Sulu Sea, South China Sea and Pacific Ocean were observed. (2)During the second mesoscale iron enrichment experiments in the western subarctic North Pacific (SEEDS-II), the effects of iron enrichment on the phytoplankton community structure and the speciation of bioactive trace metals were examined. (3)On the basis of analysis of photosynthetic proteins, Fe in seawater has been proved to be a major factor controlling the spring bloom of diatoms in the Oyashio region. (4)Diurnal dynamics of biomass of nano-flagellates and ciliates in Suruga Bay was investigated. (5)The effects of trace metals on the activity of extracellular enzymes of microbes have been proved. 4.Study on metal uptake mechanism of phytoplankton in culture (1) The size distribution of Fe in culture media of phytoplankton was investigated. Fe ligands exuded from phytoplankton were analyzed by LC-MS. (2)Membrane proteins revealed in red tide forming phytoplankton Prymnesium parvum under Fe deficiency were analyzed.
