Research Abstract |
Microtubule is one of the key machinery regulating cell cycle control, including cell growth, chromosome segregation. Although many genes and their cooperation are involved in regulation of tubulin, core component of microtubules, their functional networks are not known. Fission yeast Schizosaccharomyces pombe has played an important role for identification of cell cycle regulators and their functional analysis. Several approaches using mitotic mutant were extremely useful, and many genes were identified, and the mechanism of them became clear. We have constructed a randomly isolated mutant library of fission yeast and started systematic analysis about individual mutant genes and functional interaction among them. We have isolated 1015 strains having temperature sensitive mutation, not deletion mutation, for the purpose of analysis of most of essential genes. We have taken two kinds of different strategies to analyze tubulin regulatory network. In one approach, we investigated the genes interacting with dis1, a microtubule binding protein in fission yeast. In second approach we surveyed mutants of tubulin and their related genes. From these analysis we have found several new functional networks among them. These results are expected to elucidate the presence of multiple interactive gene networks that has not been analyzed in microtubule function.
