Research Abstract |
Tissue repair and inflammation have been well known to be an essential process made by various kinds of bioactive factors. Their failure leads to tissue remodeling followed by severe intractable diseases such as atherosclerosis and asthma. Thus, identification of factors involved in tissue remodeling and understanding of its regulatory mechanisms are considered to be very important to obviate the onset of diseases. In previous, we found that the plasma factor, histidine rich glycoprotein (HRG), participated in tissue remodeling process. In this study, we examined some biological properties of HRG including angiogenesis. In matrigel plug assay with bFGF plus heparin, HRG was shown to exert its antiangiogenic activity at 1 micromolar concentration. In particular, histidine rich domain provided the basis for antiangiogenic activity. Also, HRG was found to have significant chemotactic activity towards neutrophils. Furthermore, the expression of adhesion molecules on cell surface was shown to be regulated by some cytokines, affecting the host defense. These findings suggest that there are many bioactive substances involved in tissue repair and inflammatory process in plasma, and play an essential role under the complicated regulation.
