Research Abstract |
It was found that long-term culture of adherent murine bone marrow cells in Dexter culture medium provided us the easy way of producing large quantity of undifferentiated mesenchymal stem cell-like cells. It was also found that transfection of exogenous Ad4BP/SF-1 cDNA transformed these bone marrow-derived cells into ACTH-responsive steroidogenic cells. These cells also expressed the several genes of steroidogenic enzymes and continued to produce several steroids for more than 120 days. In order to analyze the in vivo effect of these steroidogenic cells, model mouse of adrenal failure was produced by removing both adrenal glands sequentially from scid mouse. Transplantation of the steroidogenic cells into subcapsular space of the kidney after removing one adrenal gland rescued the mice and prolonged the survival. These findings may lead to cell therapy using regenerated steroidogenic cells in the near future. This study is also significant in showing the possibility that bone marrow cells could differentiate into several lineage by changing the differentiation program of the cell, not by cell fusion.
