Research Abstract |
We conducted the comprehensive gene analysis by refusal and the DNA microarray at the time of the immunological tolerance in the mouse experiment model. In rejection, the IFN-g induction gene was strongly seen. Although the manifestation of these gene clusters maintained the immunosuppression contrary to anticipation at the time of the immunologieal tolerance, the immunological-tolerance specific candidate gene was also reported. Based on these data bases, it aimed at checking the meaning of those genes by a clinical specimen by this research. We decided to analyze a gene cluster for the analysis in an organ transplantation at the beginning. However, since the drugs of a large number besides an immunosuppressant were used in a liver transplantation and clinical backgrounds were various, analysis was difficult in practice. host and a tumor. Then, the comprehensive gene analysis of a tumor was conducted with the liver metastasis from colorectal cancer case without those with a capsule(n= 15). Under the microscope, RNA used the micro die section, extracted only oncocyte, and amplified it by PCR. About 34000 gene EST was comprehensively analyzed for this using AffymetrixU34 GeneChip. Computer software After being with GeneSpring and carrying out hierarchical clustering analysis, as a result of being with d-Chip and analyzing further, the 115-piece profile of the gene in which the manifestation is going up intentionally in a capsule(+) group as compared with a capsule(-) group was carried out. As a result of examining them in detail, the thing which set a chemokine gene cluster and to include became clear. In this, IFN-g induction chemokine in which the manifestation was going up strongly in the former Mus musculus graft rejection experiment was contained, and, moreover, elevation of a manifestation was constantly accepted by liver metastasis with capsule. In the liver metastasis in which this fact has acapsule, it is suggested that immunoreaction similar to transplant rejection has occurred between a host and cancer.
