Research Abstract |
Two to 3 / kg of bone marrow cells were obtained from iliac bone of the Beagle. Then, using Ficoll technique, mononuclear bone marrow cells were collected. These cells were cultured for approximately 4 weeks in an incubator using culture flasks consist of RPMI1640, 10% FBS and antibiotics. These cells differentiated into endothelial cells, fibroblasts, and smooth muscle cells. We fabricated small caliper biodegradable scaffold using co-polymer of PLA and PCL (P(CL/LA)) to create sponge-shaped tubular graft. PGA fibers, located at inner part of the scaffold were used for reinforcement of the scaffold. Diameter of the graft was 3.5 mm, and 5 cm in length. After 4 weeks of cultivation, culture cells were trypsinized and seeded onto the scaffold following by settle up in to the bioreactor consist of pulsatile pump, tube and oxygenator. After a few weeks cultivation in the bioreactor, the cells seeded scaffold was implanted to the same dog which the bone marrow cells were obtained. The dogs were observed for 1 and 3 months after implantation. Angiograms were performed for the evaluation. One month's graft were patent ; however, they occluded with fibrotic changes within 3 months after implantation. To overcome these phenomena, we are struggling to find better material for the small caliper tissue engineering graft.
