Outline of Annual Research Achievements

1.血液中の希少な癌細胞を検出するCTCチップ装置について基礎的性能の検討を行った.捕捉抗体として抗EpCAM抗体や抗Podoplanin抗体をCTCチップに結合させ,健常人に癌細胞株を添加した「CTCモデル」を流して血液中の腫瘍細胞捕捉及び捕捉した細胞の検出に関する検討を行い,捕捉抗体濃度などの至適条件を得た.
2.得られたCTCについて遺伝子学的解析のための基礎検討を行った.肺癌細胞株及び肺癌組織検体からDNAを抽出し,次世代シークエンサーにて癌特異的遺伝子変異を調べた.
3.高導電率溶液中の循環腫瘍細胞の単離と,誘電泳動特性の測定が可能なピラー電極アレイデバイスを試作し,そのデバイスの有用性を基礎実験により評価した.
4.循環腫瘍細胞もしくは血液細胞の1個のインピーダンスを広帯域に測定できるマイクロストリップ上平行電極型のリング共振器を設計・試作し,マイクロ粒子の有無によりデバイスの共振周波数が変化することを確認できた.また,負性抵抗素子を上記デバイスに付加することにより,高感度化(共振特性の高Q値化)を実現した.
5.誘電泳動下で回転している細胞が形状変化をすると,その回転速度が影響を受ける.誘電泳動下で回転する肺癌細胞の形状分類を行い,その分類結果と回転速度との関係を評価した.形状分類に当たっては,細胞の大きさのみならず,細胞表面の凹凸や円形度も考慮するために,自己組織化マップを拡張した方法を用いた.その結果,細胞の大きさおよび表面の滑らかさが回転速度と密接に関係することが示された.
6.クリークギャップ電極によって1個の細胞に生じる誘電泳動力の測定結果と,誘電泳動溶液内で自由落下する時のターミナル速度から求めたその細胞1個の比重から,自己組織化ファジィシステムを用いて,その細胞が白血病細胞,循環腫瘍細胞,正常白血球のいずれに属するかを同定出来た.
