KeywordsT cell receptor / Antigen / High-throughput / Method
Outline of Annual Research Achievements

In this fiscal year, I have done these experiments,
I have cultured two different types of antigen presenting cells (APC), which can express different antigens. I have cultured two different types of T cell lines, which can recognize those two APCs respectively.
I have mixed and co-cultured the T cell line and APC, and found the clustered cells.
I have tested the protocol for the construction of the plasmid with random sequences.

Current Status of Research Progress
Current Status of Research Progress

2: Research has progressed on the whole more than it was originally planned.

Reason

I have changed some parts of the original plan of the last fiscal year, however I have succeeded with the new strategy. Overall, the progress is going well, it is basically consistent with the previous expected results.

Strategy for Future Research Activity

In the next fiscal year, I will do these steps:
(1) Setup a platform for scanning the recognized T-cell-APC(Antigen presenting cell) pairs.
(2) Setup a platform for pick up these recognized T-cell-APC pairs.
(3) Do next-generation sequencing to identify the T cell receptor's sequence and the antigen sequence within pair.
(4) Write a pipeline for data analysis.
