Identification of ingredients for suppressing HIV-1 replication by type-1 gamma-delta T cells

Outline of Annual Research Achievements

これまでの研究で、Vγ1Vδ1型のT細胞レセプター(TCR)を発現したγδT細胞株(clone 1C116)の樹立に成功し、このT細胞がTCR特異的な抗原分子に遭遇した場合、IL-2を放出することを報告してきた。そして、このような抗原分子として天然生薬中に存在するフラボノイド、テルペノイド、アルカロイドなどの影響を調べたところ、その一部が応答することを見出した。本年度は、この事実を基に更に追求を重ねた結果、1C116株が糖質と結合したフラボノイド、2糖類と結合したフラボノイドであるヘスペリジンあるいはリナリンと特異的に応答することを見出した。そして、これらヘスペリジンあるいはリナリンで末梢血細胞を刺激したところVγ1Vδ1型γδ細胞が活性化され、IL-5並びにIL-13のようなサイトカイン、そしてケモカインであるMIP-1α、MIP-1β、RANTESなどのケモカインが放出されることを確認した。このような事実を基に、ヒト末梢血より純化採取したVγ1Vδ1型γδ細胞とR5-型HIV-1を感染させたCD4陽性NKT細胞を共培養した実験系において、ウイルス増殖が全く抑制されない状況下おいて、ヘスペリジンあるいはリナリンを添加した場合、著明なR5-型HIV-1の増殖抑制が認められることを発見した。ヘスペリジンは生薬の陳皮にリナリンは枸杞子の主成分であることから、生薬中に含まれる2糖類と結合したフラボノイドが、Vγ1Vδ1型γδ細胞を活性化した場合、活性化されたγδ細胞より放出されたサイトカインあるいはケモカインによって、NKT細胞内のR5-型HIV-1の増殖が抑制されることが明らかとなった。また、活性化されたγδ細胞は、CD4陽性T細胞内のR5-型HIV-1の増殖も抑制することを確認した。以上の事項をScentific Repotrs誌に投稿し、無事acceptされた。
