Outline of Annual Research Achievements

これまでの研究により、Shati/Nat8lのKOマウスでは雌性でのみ認知記憶の障害がおこることが確認され、またその障害は、海馬CA3領域でのShati/Nat8l過剰発現により、回復することを報告した。本年度はShati KO海馬CA1領域での長期増強(long-term potentiation; LTP)を評価し、Shakti/Nat8lとシナプス可塑性の関係について検討した。
LTPの検討には、MED64システムによる多電極細胞外電位測定を行った。刺激電流強度は最大応答の40%の電圧となる刺激電流の大きさとした。シータバースト刺激前5分間の平均を基準として、刺激後120分間の測定を行った。その結果、雌雄のShati-KOとも、それぞれのWTと比較して刺激後の電位の変化が低下しており、LTPが有意に障害されていた。さらに、雌雄のShati KOマウスの海馬CA3領域にShati/Nat8lを組込んだAAVベクター (AAV-Shati) を注入した。GFPのみを組込んだAAV を注入したShati KOマウスをmock群としそのLTPに対する影響を検討したところ、海馬CA3へのAAV注入は注入領域のCA3およびCA1領域でも陽性細胞が観察され、Shati-KOで見られたLTPの障害はAAV-Shatiの雌性Shati-KO海馬CA3領域への注入によって、有意に回復した。Shati/Nat8lの欠損は、LTPの障害を誘発し、特に海馬CA1およびCA3領域のShati/Nat8lが重要であることが示唆された。雌雄のShati KOで海馬のLTPが障害され、行動実験における記憶障害は雌性マウスのみ観察されたことについては、今後の検討が必要である。Shati/Nat8l、NAAまたはNAAGが新たな認知症治療薬の開発標的となることが期待される。
