Research Abstract |
Hematological malignancies include acute myeloid leukemia (AML), adult T-cell leukemia (ATL), myelodysplastic syndromes (MDS), chronic myeloid leukemia (CML), etc. There are only few disorders for which we have reliable diagnostic molecular markers. To identify such diagnostic genes, it would be useful to analyze gene expression profiles among fresh clinical specimens of the disorders by using genomics tools (such as DNA microarray). Here we have purified CD133-positive hematopoietic stem cell fractions from bone marrow of individuals with various hematological malignancies, and determined gene expression profiles of such isolated materials by using DNA microarray. Also, we purified CD4-positive helper T-cell fractions out of peripheral blood of patients with ATL. We could identify a number of genes, expression of which is linked to long-term prognosis of the AML patients, and is linked to stage progression of ATL. Furthermore, we could identify a growth factor receptor gene whose expression is directly involved in the stage progression mechanism of ATL.
