Research Abstract |
Arabidopsis TERMINAL FLOWER 1 (TFL1) is required for the establishment and maintenance of the inflorescence meristem (IM). Although TFL1 function is required in the entire shoot apical meristem (SAM), TFL1 is expressed only in the central region of the SAM. Here, we show that TFL1 protein moves from the inner region to outer layers of the SAM in accordance with the vegetative-toreproductive phase transition that is the molecular basis of the non-cellautonomous function of TFL1. This protein trafficking also occurs from the outer to inner layers of the inflorescence meristem (IM), but TFL1 protein trafficking was not observed in other tissues, such as the flower stalk, mature floral organs, or leaves. We found that TFL1 protein is required in the entire SAM by using an immobilized TFL1 protein, showing that TFL1 is responsible for cell-to-cell communication during IM development. Molecular dissection of TFL1 protein revealed that a 21 amino acid region encoded by exon 2 is essential and sufficient for TFL1 protein trafficking. Finally, we found that a paralog of TFL1, FLOWERING LOCUS T (FT), that is a floral pathway integrator involved in leaf to SAM systemic flowering signal transduction, also moves cell to cell in the SAM.
