Research Abstract

群体性ボルボックス目は,同型配偶から異型配偶/卵生殖に至る進化を研究するためのモテル系統群である。16細胞群体性Gonium pectoraleでは,+型/-型両方の交配型の配偶子が両方向的接合突起を形成するのに対し,Chlamydomonas reinhardtiiの配偶子は+型のみが片方向的に接合突起を形成し,異型配偶化の前段階で片方向から両方向へ突起の進化が推定されている。C.reinhardtiiでは,「性染色体領域」mating type(MT)Iocusが知られており,接合突起に関連した性特異的遺伝子が局在している。我々はG.pectorale MT locusのゲノム構造を明らかにし,接合突起の両方向化にともなうMT locusの変化を解明することを目指している。本年度はC.reinhardtiiで-型MT locus上の性決定遺伝子CrMIDのG.pectoraleオーソログ(GpMID)を起点としたinverse PCRによって,G.pectorale-型ゲノム上に,C.reinhardtiiの機能未知性特異的遺伝子MTD1オーソログ(GpMTDI)を発見した。GpMTDIは群体性ボルボックス目においてMID以外で見出された初めての性特異的遺伝子てあり,-型のゲノムにシングルコピーデコードされており,-型の交配型のみに遺伝した。mRNA量は配偶子特異的に増大した。BLASTXによって,GpMTDIと核移行タンパク質ファミリーkaryopherin/importinの弱い相同性が見出された。GpMTDIはMT-配偶子でGpMIDまたはその他の因子を各へと運搬する機能に関与していることが考えられる。また,性染色体領域の解読の為,Pleodorina starriiの雌雄株,Conium pectoraleのプラス,マイナス株のBACライプラリーの構築を実施した。
