Outline of Annual Research Achievements

電気化学インピーダンス解析で得られた固体腐植ヒューミンの電気容量を、固体腐植ヒューミン依存性の嫌気性ペンタクロロフェノール脱塩素微生物反応と比較したところ、大きく異なった。電気化学的特性評価のためには、電気化学インピーダンス法の改良の必要性が示された。
シンクロトロン光によるXPS解析を含む各種スペクトル分析により、固体腐植ヒューミンの詳細化学構造解析を行った。固体腐植ヒューミンの化学構造と電子供与能は、調製方法によって異なった。電子供与能を有する固体腐植ヒューミンの有機画分は、脂肪族炭素および炭水化物構造が優占し、O=C-N、O=C-Oの部分構造が細胞外電子伝達能と関連していることが示唆された。キノン骨格が検出されない固体腐植ヒューミンにも細胞外電子伝達機能が見られたことから、細胞外電子伝達能へのキノン骨格の寄与は小さいと考えられた。
固体腐植ヒューミンに活性を依存する嫌気性ペンタクロロフェノール脱塩素細菌は、脱塩素反応に必要な電子を固体腐植ヒューミンから直接得ることが判っている。そこで、この微生物群に対して固体腐植ヒューミン還元のための化学物質として水素を供与し、ペンタクロロフェノールの脱塩素反応から、水素から固体腐植ヒューミンを還元するまでの電子の流れに関与する微生物群の群集構造解析を行った結果、Firmicutes門が優占(96%)し、続いてProteobacteria門(<3.5%)であった。Firmicutes門は、63%がTissierella科(Tissierella_Soehngenia)、30%がLachnospira科で96%の内の殆どを占めた。
