Outline of Annual Research Achievements |
本研究では、ケモカイン受容体XCR1を発現する樹状細胞サブセット(XCR1+DC)を恒常的に欠失するマウス(XCR1-DTAマウス)を用いて、XCR1+DCの機能的意義の解析を進めている。今回、移植片対宿主病(GVHD)におけるXCR1+DCの機能的意義を検討した。GVHDは、骨髄細胞と脾臓細胞を、MHCが異なる野生型マウス(レシピエント)へ移植することにより誘導した。具体的には、放射線照射した野生型マウス(レシピエント)に、X線照射したXCR1-DTAマウス由来の骨髄細胞と脾臓細胞をドナーとして移植した場合(A)と、野生型マウス由来の骨髄細胞と脾臓細胞をドナーとして移植した場合(B)の生存率を比較した。その結果、(A)の生存率が(B)よりも高く、ドナー由来のXCR1+DCがGVHDの病態に関与することが示唆された。 また、本研究では、これまでに、粘膜系免疫アジュバントであるコレラ毒素(CT)のBサブユニット(CTB)が、リポ多糖(LPS)と相乗的に、腹腔内に常在するマクロファージ(腹腔マクロファージ)から炎症性サイトカインIL-1β産生を誘導することを明らかにしていたが、さらに、CTBにより発現が誘導される遺伝子群の解析を行ったところ、小胞体ストレス応答に関与する遺伝子群の発現が誘導されることを見出した。そして、阻害剤を用いて、小胞体ストレスセンサーのうち、IRE1αがCTBによるIL-1β産生誘導に関与することを見出した。この結果から、小胞体ストレス応答が、CTBによる炎症性サイトカイン産生誘導に関与することが示唆された。
