Outline of Annual Research Achievements

<1.メチローム解読> Hp Genome Projectが世界からピロリ菌1000株を収集し、その半分のメチロームをPacbioシーケンサーで解読した。私たちは、杏林大医学部、札幌厚生病院の株を送った。
<2.地域特異性> 私たちとChinaCDCによる、東アジア各地のピロリ菌350株のゲノム配列について、系統地理解析を進めた。東アジアの先史・歴史、日本人の起源(縄文、弥生)に対応する集団構造が得られた。分集団特異的なタンパク配列多型(高Fstサイト)が多数明らかになり、立体構造ベースの機能予測から発がんとの関連を推測した。
<3.GWAS> 日本とベトナム由来の十二指腸潰瘍株と胃がん株のGWASから、この病態差に関連するタンパク配列多型を明らかにし、構造から機構を推測した。HpGP株の胃がんvs.胃炎の予備的GWASから、多数のタンパク配列多型を明らかにし、構造から機構を推測した。
<4.ミクロ進化> 配列特異的DNAメチル化酵素をハブとする、遺伝子制御ネットワークによる、適応的形質の支配を明らかにした。DNAメチル化と翻訳と転写の関連を示した。病原性アイランドでの、メチル化モチーフごとの特徴的な分布と遺伝子発現制御との関連を明らかにした。家族内感染株のメチロームの比較から、ピロリ菌メチル化酵素遺伝子の再編による配列特異性変換が、メチロームのミクロ進化を進める事を明らかにした。これらの株をスナネズミに感染させて、ゲノム進化を追った。発がんタンパク候補の機能解析を開始した。
<5.Pacbio> ピロリ菌メチロームのPacbio解読のマルチプレックス法を、Pacbio社とともに開発した。1 セルで16株分を読む。
