Outline of Annual Research Achievements

Gタンパク質共役型受容体(GPCR)は複数のエフェクタータンパク質を介して細胞内シグナル伝達を誘導する。それぞれのGPCRは異なるパターンでエフェクタータンパク質に結合することで個々のGPCRに特徴的な細胞応答が誘起される。近年の創薬研究では内因性リガンドとは異なるパターンのシグナルを誘導するGPCRリガンドが着目されている。この作用を有するGPCRリガンドはバイアスリガンドと呼ばれ、薬効の主作用のシグナルを維持したまま、副作用のシグナルを減弱させることができ、副作用を切り分けられる創薬として期待される。本研究では三量体Gタンパク質とβアレスチンのバイアスや三量体Gタンパク質のサブタイプのバイアスシグナルに着目し、GPCR作用薬のシグナル活性測定とその分子機序の解明に取り組んだ。本研究課題では前年度までに見出したAT1アンジオテンシンII(AngII)受容体のGq、Gi、G12のシグナルバイアスについて、臨床薬TRV120027(以下、TRV)をペプチド合成しそのシグナル効果を調べた。その結果、TRVのGタンパク質活性化プロファイルはAngIIと比較して、Gqが弱くGiやG12に対して強いシグナルを誘導した。また、TRV はβアレスチンのAT1への会合を誘導し、これはAngIIとは異なる挙動であった。Gq阻害剤(YM-254890)を添加した条件ではAngIIのβアレスチンのAT1への会合応答がTRVと同様になることを見出した。従って、Gq活性化はβアレスチンの制御に重要であることがわかった。さらに、Gqシグナルを解析する手法として、スプリットルシフェラーゼによりGqとPLCβの相互作用を検出するセンサーを構築した。
