Outline of Final Research Achievements |
Salivary gland tumors have diverse histology, and tumor-specific genetic alterations have recently been identified. In our study, about 50% of polymorphous adenocarcinomas had polymorphous adenocarcinoma-associated genetic abnormalities of PRKD1/2/3, ARID1A, and DDX3X rearrangements, and fluorescent in situ hybridization (FISH) revealed identical genetic abnormalities in tubular, cribriform, papillary, solid, and microcystic forms. In pleomorphic adenomas, PLAG1 rearrangements were also detected in tubular cells, myoepithelial spindle cells, myoepithelial oncocytic cells, myoepithelial plasmacytoid cells, and mesenchymal chondroid cells, but not in vascular endothelial cells.Whole-slide FISH clearly identified tumor or non-tumor cells and was effective in elucidating tumor differentiation.

Academic Significance and Societal Importance of the Research Achievements |
唾液腺腫瘍は近年、特異的な遺伝子異常が報告されている。我々の研究では、whole-slide FISHに、組織像は多彩である多形腺腫などの唾液腺腫瘍を用いることで、組織像、形態の違いがあっても腫瘍細胞には同一の遺伝子異常を有することが明らかになり、血管内皮細胞などには遺伝子異常を有しないことが示された。これにより、組織内における腫瘍部分と非腫瘍部分を明白にすることが可能であることが示唆できた。今後は、同一スライドを用いて様々な遺伝子異常を調べることで、腫瘍形成の過程を知る一助になると考えている。
