Outline of Annual Research Achievements

本研究は、国内の年縞堆積物を対象として環境DNA分析を実施して、化学成分や微化石から復元できない動植物相や人間活動履歴情報を網羅的に把握することができるのか、その検証をはじめて試みることである。
そのために、気候環境や文化人類学的に景観の異なる北海道藻琴湖および、静岡県浜名湖に堆積する完新世（過去約1万年間）の湖底堆積物（年縞）を対象として、環境DNAの解析手法を援用して、景観を構成する生物相や人間活動履歴を時系列変化として復元する。
最終年度となった平成30年度においては、その前年度に採集した北海道藻琴湖および、静岡県浜名湖の年縞堆積物の堆積学的、古生物学的研究を進めた。その一方、DNAの損傷程度の確認および、分析フローの確立を目的として、堆積年代の異なるスポットサンプルによる、メタ解析によるDNA分析を世界ではじめて実施した。
その結果、藻琴湖においては、かつて生息していたイトウの在不在を議論できる可能性が見いだされた。浜名湖においては、従来の方法にて湖水環境が一変した西暦1498年の前と後、また、太陽活動と関連する小氷期の気候変動を復元できた。しかしながら、このような環境変化における海水魚の出現頻度については、明瞭な関係を見出すことができなかった。この原因については、湖沼ごとにDNAの損傷程度が異なることが推定され、時代をさかのぼるにつれ、損傷する傾向以外に、堆積環境による損傷程度に違いがあることが示唆された。
