Research Abstract

本研究では、細胞機能発現における低分子量G蛋白質Rhoを介するシグナル経路の役割と、この経路の個体発生および病態への関与を解明することを目的とし、以下の成果を得た。(1)Rho標的蛋白質mDia1欠損マウスより得たMEF細胞に温度感受性v-Srcを感染させ、mDia1が、v-Srcの細胞辺縁への集積に寄与すること、また、この集積がSrcによる細胞悪性化、浸潤、個体での腫瘍形成に必須であること見出した(Tanji et al.)。(2)mDia1/3の二重欠損マウス(DKO)を用い、発生時の神経上皮組織においてRho-mDia経路が神経幹細胞を繋ぐapical actin beltの形成に寄与し、その破綻が神経幹細胞の無秩序な増殖・分化を促すことを見出した(投稿中)。また、DKOでは、嗅球低形成が観察され、この原因として、神経前駆細胞の接線方向への移動の障害が起こることを見出した(投稿中)。(3)mDia2結合蛋白質を探索し、anillinを同定した。mDia2のRhoおよびanillinとの結合が、収縮環局在、細胞質分裂時での機能発現に必須であることを見出した(Watanabe et al.)。(4)新規mDia結合蛋白質Liprinを見出した。Liprinは、Rho・mDia依存的なアクチンストレス線維形成を抑制すること、LiprinがRho依存的なmDiaの細胞膜分画に移行を抑制することを見出した(in revision)。(5)ROCK-I、-IIダブルヘテロマウスの交配を行い、DKOは胎生3.5~9.5日で致死に、ホモ・ヘテロ/ヘテロ・ホモ欠損マウスは、胎生9.5日でbody turningやyolk sacでの血管発達不全を呈することを見出した(投稿中)。(6)ヒトES細胞分散培養時にはRho-ROCK系の活性化に加え、Rac活性が減衰すること、また、これら経路はAbrにより制御されていることを見出した(Ohgushi et al)。(7)単分子蛍光偏光によりmDia1の可視化を行い、アクチン線維の螺旋周期構造に一致してその偏光の向きが振動することを見出した(Mizuno et al)。
