新規脳分子による新しい生殖制御機構の解明

Research Abstract

我々は生殖腺刺激ホルモンの放出を抑制する新規視床下部ペプチドを鳥類から同定して生殖腺刺激ホルモン放出抑制ホルモン(gonadotropin-inhibitory hormone; GnlH)と名付けた。昨年度までの研究により、GnlH同族ペプチドを霊長類、哺乳類、爬虫類、両生類、魚類、無顎類などから同定して、GnlHとGnlH同族ペプチドの起源と構造の分子進化の大略を明らかにした。GnlHとGnlH同族ペプチドはC-末端側の配列がいずれも共通のLeu-Pro-Xaa-Arg-Phe-NH2(Xaa=Leu or GIn)(LPXRFa)構造を持つという特徴があり、GnlHとGnlH同族ペプチドの起源は少なくとも無顎類に遡ることがわかった。本年度は、GnlHとGnlH同族ペプチドの生殖腺の発達と機能における生理作用と作用機構を主に哺乳類や鳥類などの高等脊椎動物を用いて解析した。その結果、GnlHとGnlH同族ペプチドは下垂体に存在する新規のGタンパク共役型受容体を介して生殖腺刺激ホルモンの合成と放出を抑制して、生殖腺の発達と機能を抑えることが明らかになった。また、GnlHとGnlH同族ペプチドの受容体は生殖腺刺激ホルモン放出ホルモン(gonadotropin-reIeasing hormone; GnRH)ニューロンにも存在しており、GnlHとGnlH同族ペプチドはGnRHニューロンに作用して生殖腺刺激ホルモンの合成と放出を抑制することが見いだされた。さらに、GnlHとGnlH同族ペプチドは生殖腺にも発現しており、GnlHとGnlH同族ペプチドの局所作用により、配偶子形成が抑制されることが示唆された。GnlHとGnlH同族ペプチドの発現制御機構を解析したところ、メラトニンがGnlHとGnlH同族ペプチドの発現を誘導して生殖を抑制することが明らかになった。
