Research Abstract

妊娠ラットにTBTを0, 25, 50, 125ppm含む餌を摂取させて、生まれたオスのF1ラットを生後1、2、3週の時点で処理をした。更に餌を通常の餌に切り替え、6週の時点でも処理を行った。ラットの脳からRNA抽出を行い、RNAからアミノアリル基を導入したcDNAを合成し、オリゴヌクレオチドマイクロアレイに適用した。対照群と曝露群の間で1.5倍以上の変化があった遺伝子に関し、リアルタイムPCRを用いて定量的に解析した。F1ラットの中脳でのmRNA発現について、ミトコンドリアの呼吸鎖複合体4におけるシトクロムc酸化酵素の発現上昇、複合体1に関する遺伝子の発現変化、活性酸素分解酵素の発現変化、神経伝達物質の貯蔵に関する遺伝子の発現上昇が観察された。以上より、TBTによるF1ラットの発達神経毒性は曝露後にも更に顕著になる可能性が示された
