Outline of Annual Research Achievements

研究初年度である2018年度は,汚染空気暴露に伴う健康影響を可視化するin silico人体モデル開発に向けて,以下の課題に取り組んだ.
(1) In silico人体モデル開発のための人体プラットフォーム作成:人体詳細幾何形状を再現したCFD解析用の数値人体モデルと鼻腔から気管支第四分岐までを再現した数値気道モデルを基に,人体皮膚表面幾何形状と呼吸器系幾何形状を精緻に再現したin silico人体モデルのプロトタイプモデルを作成した.これはCFD解析の人体幾何形状メッシュであると共に,部位別の生理的薬物動態モデルや体温調節モデルを統合するためのプラットフォームとなる.
(2) 経気道暴露用の生理的薬物動態(PBPK)モデルの作成:鼻腔・口腔から気管支までの上気道を対象とした生理的薬物動態モデル(経気道暴露用PBPKモデル)の定式化を行った.本年度は,気道界面の粘液,粘膜上皮,上皮下組織からなる細胞組織層を再現し,各層で代謝,非特異反応・結合,組織内拡散,血流輸送を記述する反応・拡散系数理モデルを基礎とするPBPKモデルを提案し,計算速度の検討,パラメータの感度解析など基礎的な検討を行った.
(3) 経皮暴露用の生理的薬物動態(PBPK)モデルの作成:室内空気環境と皮膚界面の境界条件として,表皮,真皮,皮下組織からなる経皮曝露用PBPKモデルを定式化した.表皮に沈着した汚染物質は表皮内での代謝,非特異反応・結合の他,組織内を拡散移動するモデル化とし,皮下組織内に到達後は反応・拡散に加えて血流により体内動態する様相を反応拡散系数理モデルとして定式化した.予備解析として環境煙草煙に含まれる各種化学物質を対象として経皮暴露シミュレーションを実施した.

Strategy for Future Research Activity

研究計画に従い,実直に研究を推進する.現時点で研究計画変更の必要性や大きな問題点は無いが,本年度に作成したin silico人体モデルとCFDの連成解析に要する数値解析用メッシュ数が1000万メッシュを超え,計算負荷が非常に大きくなっている.この問題に対し,計算アルゴリズムの見直しなどで計算速度の改善を検討すると共に,クラスタ計算機を導入することでも計算速度の改善を検討する予定である.
