| Outline of Annual Research Achievements |
192個体分のヒトゲノムDNAを本年度の解析対象試料とした。嗅覚受容体（OR）、苦味受容体（TAS2R）、旨味甘味受容体（TAS1R）、L・M・Sオプシン、noncoding中立対照のゲノム領域をtarget captureと大規模並列シーケンシングの対象とした。ORについては554遺伝子をプローブ作成対象とし、それらの内訳はヒト標準ゲノムデータベースhg38からintact ORを398個、hg38中の標準ゲノム配列に含まれないaltハプロタイプを4個、hg38の標準ゲノム配列上にあるOR偽遺伝子の内ORクラス1/2それぞれのアラインメント上で保存性の高いサイトの内ギャップ数が3個以上5個以下のものを4個、同じくその内first Metから終止コドンまでの間にinterrupting stop codonかフレームシフトのどちらか一方を1個だけ含み上記高度保サイトにギャップ数が2個以下のものを91個、同じくfirst Metから終止コドンまでの間にinterrupting stop codonかフレームシフトのどちらか一方を1個だけ含み上記高度保サイトにギャップ数が3個以上5個以下のものを4個、チンパンジーゲノムから同定された397個のintact OR genesのうちチンパンジーにあってヒトにないものを53個とした。TAS2Rについてはヒト上科共通祖先時点でintactと推定されるヒト29遺伝子、中立対照についてはヒトゲノム非遺伝子領域から単一コピー156領域をプローブ作成対象とした。
葉食性のコロブス類について行動実験と組み合わせた結果、苦味受容体（TAS2R38）についても甘味（TAS1R2/TAS1R3）についても機能が減弱していることがわかった。このことは、コロブス類が葉食性に特化していることと関連があることが示唆される。また、他の受容体（TAS2R10,16,14,46）についても機能解析系を立ち上げた。

196個体分のヒトゲノムからtarget captureと大規模並列シーケンシングを実施し、これまでの210個体分と合わせて配列解析を開始した。アジア・アフリカのサル9種に関して、TAS2R遺伝子のtarget captureと大規模並列シーケンシングによる配列解析を開始した。中南米のサル10種に関してOR遺伝子のtarget captureと大規模並列シーケンシングによる配列解析を前進させた。中南米のサル、アジア・アフリカのサル、類人猿、ヒトにおけるL-Mオプシン遺伝子間塩基相違度解析を前進させた。

テナガザルについてこれまでの3種に加え6種、中南米のサルについてこれまでの10種に加え9種、メガネザル1種、曲鼻猿9種（キツネザル類4種、ロリス類5種）について嗅覚受容体（OR）、苦味受容体（TAS2R）、旨味甘味受容体（TAS1R）、L・M・Sオプシン、noncoding中立対照のゲノム領域のtarget captureと大規模並列シーケンシングを実施する。ヒト402個体分についてORのtarget captureと大規模並列シーケンシングによる配列解析を開始する。テナガザル9種についてTAS2R遺伝子のtarget captureと大規模並列シーケンシングによる配列解析を開始する。アジア・アフリカのサル9種についてTAS2R遺伝子のtarget captureと大規模並列シーケンシングによる配列解析を進展させる。中南米のサル19種についてOR遺伝子のtarget captureと大規模並列シーケンシングによる解析を進展させる。中南米のサル、アジア・アフリカのサル、類人猿、ヒトにおけるL-Mオプシン遺伝子間塩基相違度解析をさらに前進させる。
