Outline of Annual Research Achievements

腸管粘膜は、食餌性成分や腸内細菌叢の変動を可及的速やかに感知し、変化やストレスに応じて適応することで組織恒常性を維持する。しかしながら、その機能が破綻することで炎症の遷延化が起こり、炎症性腸疾患の発症や再燃、増悪化につながる。
クローン病では、腸管の蠕動運動や形態形成を司る粘膜下支持組織に至るまでの腸管全層において炎症反応や機能不全が起こる。粘膜炎症を起点とし、炎症因子もしくは腸内細菌成分に対して感受性がより高い粘膜下もしくは筋層の構成細胞に作用し、炎症反応が加速する。このような負の連関の作用機序の解明に向け本年解析を進めた。さらに、accessory digestive organ(腸管機能支持組織)として位置付けられる「膵臓」と「腸管」との「腸-膵連関」による腸管粘膜の恒常性構築・維持機構が、抗膵臓自己抗体の産生によって遮断される。その結果、炎症の増悪化が起こることが示された。
これらの連関システムの破綻モデル動物を作製し、連関の統合的な解析を進め、病態形成に関わる腸内細菌を同定した。これは、接着性浸潤性大腸菌とは異なる新たな炎症起因菌の候補菌である。
上記に加えて、「粘膜―間葉系連関」並びに「粘膜-筋層連関」プロジェクトにおいては、マウス筋層の単細胞解析を実施し、蠕動運動にかかわる細胞集団の分類、さらには常在菌シグナルの有無や炎症ストレスによって起こる構成細胞の変化並びに機能的変遷についての解析を進めた。
