植物の間接防衛の誘導機構解明と防除への応用

Research Abstract

シロイヌナズナの野生型およびHPL活性の無いミュータントを用いて緑の香り(て青葉アルコール、青葉アセテート、青葉アルデヒド)が媒介する植物間のコミュニケーションの感度を測定した。その結果、感度は概ね140pptVの緑のかおりに反応できるほどの高さであることを明らかにした。さらに植物間コミュニケーションを利用した害虫防除への応用にむけた実証試験をダイズ圃場で行い、良好な結果を得ている。
トマトで見いだされたヘキセニル配糖体の構造を確定したことを受けて、昨年に引き続き得られたヘキセニル配糖体がハスモンヨトウ成長を阻害することを明らかにした。今年度はこうした取り込み、変換がヘキセノール特異的であることを確認するためさまざまな構造類縁体でトマト無傷植物を処理し、HPLC-MSにより配糖体化特異性を解析した結果、一部の化合物で特異性が認められた。また、配糖体化酵素の同定を引き続いて行なっている。曝露植物でマイクロアレイを実施し、誘導される遺伝子を同定した。
除虫菊は昆虫防御物質として天然殺虫剤ピレトリンを生合成する。ピレトリンは非メバロン酸経路でつくられる酸部とオキシリピン経路でつくられるアルコールとがエステル結合することで生じる。このエステル結合の形成を触媒するアシルトランスフェラーゼを除虫菊の蕾から世界に先駆けて精製し、遺伝子をクローニングしたところ、驚くべきことに酵素はGDSLリパーゼであった。本酵素は基質の菊酸CoAとピレスロロンの絶対配置を厳密に認識した。また、EGFP融合体の挙動を調べたところ酵素は細胞外に分泌され、細胞壁と原形質膜の間に蓄積することが判明した。植物各組織での本酵素遺伝子の発現量はピレトリン含量とよく一致し、遺伝子は傷害に応答して発現したことか、本酵素は除虫菊の対昆虫の防御に重要な役割を果たしていることが示唆された。
