Research Abstract

ヒトゲノム計画の成果により、患者の遺伝的体質に合わせた処方・治療計画を行う、いわゆるテーラーメード医療が現実のものになろうとしている。遺伝子発現の量的変動に関する包括的情報を活用し、テーラーメード医療を実現するには、遺伝子発現情報と臨床情報を統合することが必要である。H19繰越基盤研究(S)に於いては、基盤研究(S)研究全体で用いる統合遺伝子発現解析プラットフォームの方法論の選定を行った。
目的:マイクロRNAマイクロアレイを将来、臨床診断に用いようとするならば、MicroArray Quality Control(MAQC)プロジェクトがmRNAマイクロアレイの再現性を確かめたように(Nature Biotech,2006)、マイクロRNAマイクロアレイの再現性の検討が必要である。そこで、我々は市販されている5つのマイクロRNAマイクロアレイの再現性を検討した。
方法:マイクロアレイのプラットフォームとしてアジレント社、アンビオン社、エキシコン社、インビトロジェン社、東レ社を選んだ。RNA標本として、アンビオン社のヒト肝臓、ヒト前立腺組織total RNAを用い、各社推奨の実験プロトコールに従って、3回づつハイブリ実験を行った。
5つのプラットフォームに共通な309種類のマイクロRNAに関してその再現性を検討した。
結果と考察:東レ・アジレント社のマイクロRNAマイクロアレイは、同一プラットフォーム内の再現性が高く、将来の臨床診断に応用できる可能性を示した。
マイクロRNAマイクロアレイ法は、現在のところ、異種プラットフォーム間でのデータの再現性については高いとは言えないが、同一プラットフォーム内でのデータの再現性は高いと考えられる。今回使用したプラットフォームでは、東レとアジレントのプラットフォームが高い性能を示した。
