Research Abstract

微生物ガス代謝とアイソトポマー比の関係について、純粋培養系と現場環境観測で得られる知見のギャップを埋めるべく、いくつかの微生物代謝過程に注目し、より現場環境に近い微生物共存・競合培養実験系(複数の微生物過程が共役したり競合したりする系)を考案し、微量アイソトポマー比測定技術を駆使することで、実験で発生するガス種および基質のアイソトポマー比計測を実現することを目的として研究を行っている。本培養(濃度・アイソトポマー比測定実験)を行うために、ジャーファーメンター、pH・溶存酸素・栄養塩濃度調節器、送液ポンプなどから成る連続培養装置を設置し、運転条件や気体・液体試料の採取方法について検討した。定常状態で系内のCH_4、CO_2、H_2S、SO_4の定量を行う方法を構築した。H_2Sに関しては四種硫黄同位体分析の高精度化のため、硫化銀のフッ化によるSF_6法を改良した。フッ化およびSF_6ガスの精製の前処理装置により、すでに<33>^S/<32>^S比の測定は本研究目的に十分な精度(±0.01‰)で分析が可能であった。一方<36>^S/<32>^S比は±0.5‰の精度で分析可能であるが、現段階で十分な精度(<±0.2‰)に達していなかった。多段階ガスクロマトグラフィーを新たに適用して、まずSF_6ガスの精製法を改善し四種硫黄同位体分析法の高精度化を達成した。純水製造装置と、新たに設置したターボポンプにより、水質と不純ガスの排除を保障した系で微生物過程により放出された各種成分を新たに購入したGCMSにより定量したのち、アイソトポマー比を現有の同位体比計測用質量分析計で計測した。得られた結果を解析し、微生物ガス代謝とアイソトポマー比の関係を関数化するとともに、実験系の評価と再設計を行った。
