Research Abstract |
申請者らは、糖尿病血管傷害の鍵となる分子が細胞膜の糖化タンパク受容体RAGE(receptor for advanced glycation endproducts)であることを明らかにするとともに、選択的スプライシングにより生成する分泌型RAGEタンパクを発見した。分泌型RAGEはデコイ受容体として機能し血管保護作用をもつと推定される。本研究計画は、分泌型RAGE過剰発現マウスと分泌型RAGEを発現しないマウスの両方を作製し、糖尿病をはじめとする各種病態における分泌型RAGEの重要性を検証するとともに、分泌型RAGE生成にいたるスプライシング機構の解明と、当該スプライシングへの薬剤効果を評価するハイスループットスクリーニング系の開発を基に、分泌型RAGEの発現制御に基づく新たな糖尿病合併症の予防・治療薬の探索を目指すものである。
