| Research Abstract |
ピリンタンパク質は4つの主要なタンパク質ドメインから構成され、同様にピリンドメインを持つファミリ-タンパク質と相互作用し、炎症性サイトカインであるIL-1βの産生を抑制し、さらにNFκBの活性化を抑制し、炎症を終息させる働きを持つ。このピリン蛋白の中でヘリックス構造をなすピリンドメインは蛋白-蛋白の相互作用に関わる重要な部分である。我々はピリンドメインの相互作用するプラットフォ-ム形成を阻害することによって炎症の抑制を行うため、細胞内導入ペプチドシークエンス(KKKKKKKKK)をアミノ末端に付加し、さらにカルボキシ末端にはHAタグシークエンスを付加したピリン蛋白由来のピリンドメインを大腸菌で作成した。この蛋白をNiカラム、ゲル濾過、陰イオン交換カラム、そしてエンドトキシンを除くためにポリミキシンカラムで精製し、マウス気管支喘息モデルを作成後、経鼻的に導入しその効果を検討した。結果 : 血清、BALF中の炎症性サイトカインを検討し、IL-5がピリンドメイン導入した群で有意に低下し、好酸球の減少も確認された。また組織の検討ではピリンドメインタンパク質は気道上皮細胞、気道上皮下の単核球にもその導入が確認された。これらの結果は経気道的投与により、吸入ステロイド薬に替わり選択的にNFκBを抑制しアレルギー性気道炎症を抑える治療薬の可能性を示した。
