Research Abstract |
多発性骨髄腫の病型診断と治療法の開発に寄与するバイオマーカーを同定することを目的として今回の研究を行つた。そのために, 多色蛍光染色体分析(SKY)法とゲノムアレイ解析により, 発症と進展に関与する遺伝子異常の同定に注力した。多発性骨髄腫とB細胞リンパ腫のゲノム異常を比較検討し, 両者に共通し高頻度に異常を検出した領域の一つである1lq22のlossと18q21. 1-q21. 3のgainを解析した, 11q22領域のlossは4Mbであつたが, 候補遺伝子はまだ同定できていない。18q21. 1-q213領域に存在するSM2の発現をRTPCRで解析した。その結果, D釘の発現に明らかな差が認められた。B細胞AD4, DCC, STARD6, TDF4, TXNL1, WDR7, FECH, NARS, MALT1, BCL2の発現をRT-PCRで解析した. その結果, DCCの発現に明らかな差が認められた. B細胞リンパ腫株では発現が消失するか非常に減弱していた。一方, 多発性骨髄腫では殆どに発現が認められたが, 3株でexOnlを欠いた発現産物を検出した。Exonlにかわつて結合する未知の配列の可能性について, Bubble PCRで検討中であ
