Repair of Oxidative DNA damage in xeroderma pigmentosum (XP) cells and its relation to XP phenotype

Research Abstract

本邦色素性乾皮症(XP)患者の約半数に合併し、患者予後に深く関わる神経学的異常の発症メカニズムは未だ不明である。今回、in vitroでルシフェラーゼ発現ベクター(pLuc)にローズベンガル、可視光線処理して光力学反応によりプラスミドDNA内に酸化的DNA損傷を誘発し、その損傷の修復力を宿主細胞回復能を指標にして解析した。材料としては研究代表者が長年所持している各群XP細胞(XPA細胞10系統、非XPA細胞22系統)を用いた。その結果XPA群細胞では紫外線性DNA損傷の修復に重要な機能を果たしているヌクレオチド除去修復能のみならず塩基除去修復系にも異常があることが判明した。XPA群以外のXP細胞においても一部で修復能の低下が見られた。この結果は酸素を最も消費する部位である脳神経細胞内に残存する酸化的DNA損傷がXP神経症状の発症に関わっている可能性を示唆する。
