Research Abstract |
HRD1の発現抑制は,APPとAβの増加をもたらすとともに,小胞体ストレスと神経細胞死を誘発することを示めし,AD患者の大脳皮質において,HRD1タンパク質が低下していることを明らかにした.さらに,APPの分解に関与する新規エビキチンリガーゼも新たに見出した.また,小胞体ストレス応答転写因子ATF6とXBP1がAPPの分解を促進し,Aβの産生を減少させることを明らかにした.さらに,HRD1やSEL1, Kf-1を含むERAD分子は,脳神経細胞に発現していることを示した.
