Outline of Annual Research Achievements

【研究項目2 次世代シークエンシングによる蚊ゲノムの解読】
217個体のネッタイシマカ蚊成虫から個体ごとにDNAを抽出し,次世代シークエンシンサーを活用したDouble Digest Restriction site Associated DNA Sequence(ddRAD-seq)を行った。ddRAD-seq解析の前に、まずネッタイシマカのゲノム情報を得るために最適なddRAD-seqのライブラリ調整条件(制限酵素の選択、PCR条件など)を見つけた。その最適な条件に基づいてライブラリを調整し、次世代シーケンサーIllumina Hiseqでゲノム全体に分布する膨大な数の一塩基多型(SNP)を約1万領域見つけた。このddRAD-seqでは、10-30個体のDNAをプールして解析を行うことで、より多くの個体のゲノムワイド配列データを迅速に取得することに成功した。

【研究項目3 デング熱媒介蚊の生息分布と個体数の拡大過程の遺伝学的推定と環境履歴との関係解析】
中立SNP領域の配列データから,分子時計で推定した進化系統樹を都市ごとに作成する。各都市の気候・洪水・土地利用等の環境履歴に基づき,機械学習モデル(ランダムフォレスト)で蚊個体数や生息の有無に影響した各都市の環境条件を解明した。主に、降水量と相対湿度と低層住宅地の密度が蚊の生息に影響していることがわかった。また、SNPを使って都市内の地域間交流の強さを遺伝子頻度の類似性に基づいて評価した結果、雌と雄の飛翔パターンに大きな違いはないことが明らかにされた。また、大きな幹線道路をまたぐ方向の蚊の移動が制限されていることも示唆された。
