Research Abstract

生体は,食物および酸素の内包する毒性やそれらの過不足といった環境ストレスに対して,応答して恒常性を維持している.本研究では,環境応答転写制御におけるストレスセンサーの分子機構を明らかにする事を目的としている
Nrf2-Keap1システムによる親電子性物質・酸化ストレス感知機構の解析:本年度は,ストレスセンサーとして働くKeap1-Nrf2複合体の結晶構造解析を目的として,これら分子を大腸菌発現系にて発現・精製した.現在,結晶化の条件を検討している.動物個体レベルの解析として,Keap1あるいは,その変異体を発現するトランスジェニックマウスを作製し,Keap1欠損マウスの表現型のレスキュー解析を行った.その結果,Keap1の複数のシステイン残基が,親電子性試薬の応答に必須であり,それぞれ性質が異なることが明らかとなった.
Epo遺伝子制御機構の解析よる低酸素感知機構の解析:Epo遺伝子の制御機構の解析を解析することで,腎臓での新たな低酸素応答の分子機構を明らかにすることを目的とし.今年度は,同遺伝子制御領域を含む大腸菌人工染色体(BAC)に緑色蛍光蛋白質(GFP)遺伝子を組み込み,レポータートランスジェニックマウスを作製した.その結果,Epo遺伝子転写制御領域に,これまで知られている低酸素応答領域とは別の,腎臓特異的・低酸素応答性の転写制御領域が存在することを明らか,その領域の範囲を2kbpにまで狭めることに成功した.
