Research Abstract

生体は,食物および酸素の内包する毒性やそれらの過不足に由来する環境ストレスに対して,適切に応答して恒常性を維持している。本研究は,環境に応答して転写制御が行われる際に、ストレスを感知するセンサーの分子機構を明らかにすることを目的としている。
Nrf2-Keap1システムによるストレス感知機構および同システムと発癌との関連性の解析:オートファジー系の異常により分解されずに蓄積したp62が、Keap1蛋白質のNrf2結合部位に競合的に結合し、Nrf2の恒常的活性化を惹起することを明らかにした。このことは、Keap1遺伝子やNrf2遺伝子自身に変異が存在しなくても、Keap1によるNrf2の活性化調節機構の破綻が、癌化、癌悪性化の誘因となる可能性を示唆している。発癌予防や癌治療に向けて、Keap1-Nrf2システムを標的とした臨床開発が期待される。また、様々ながん細胞において、Keap1-Nrf2系が破綻していることを明らかにし、癌細胞の生存戦略の新たなモデルを提唱した。
エリスロポエチン(Epo)遺伝子制御機構と低酸素感知機構の解析:腎臓でのEpo遺伝子発現は成体の低酸素応答で極めて重要である。本研究では、Epo遺伝子の発現制御機構を詳細に解析することで,新たな低酸素応答の実体を明らかにすることを目的としているが、本年度は、同遺伝子の腎臓特異的なエンハンサーとして同定した領域には、複数の制御シスエレメントが存在することを明らかにし、また、このエンハンサー以外にもEpo遺伝子の腎臓特異的な発現を支持する制御領域が存在することを示した。現在、その詳細を解析している。
