Outline of Final Research Achievements |
A squalene consuming high-throughput selection system was optimized and applied to the BmeTC mutant Y167A/D373C. After screening, we succeeded in obtaining three mutants that biosynthesize ambrein, a useful natural product, four to six times.

Academic Significance and Societal Importance of the Research Achievements |
スクアレン消費型ハイスループット選抜系はBmeTC変異体において適用すると高い致死性を示してしまい、ライブラリーの構築が困難であった。様々な条件検討を行いBmeTC変異体における最適な条件を見出した。結果、希少天然物であるアンブレインの生産性が高い変異体が3つ得られた。本研究が成功すれば、将来、全ての有用物質の大量生産が化学合成から生物合成へ置き換わる日が近いものと考えられる。未来を創造する先駆的な研究に位置づけられることが期待される。
