Outline of Final Research Achievements

Macrophage galactose-type C-type lection positive (MGL+) cells distributed in the upper dermis, especially around small vessels. In both Psoriasis (Pso) and Atopic dermatitis (AD) skin, the distribution pattern was not different from healthy control(HCs), however the number and density of MGL+ cells in the dermis and epidermis was significantly greater than in HCs. The relationship between the number of MGL+ cells and the serum levels of TARC/CCL17 and total IgE, the number of TARC/CCL17+ cells in the skin. In AD patients, positive correlations were found between the number of MGL+ cells in the dermis and the serum levels of TARC/CCL17, total IgE, and the number of TARC+ cells in the dermis. Double-staining with antibodies specific for innate immune cells revealed that the major MGL+ cell population was CD1c+, but few CD1a+, CD14+, and CD68+ cells were also observed.

Academic Significance and Societal Importance of the Research Achievements

ヒト皮膚に分布するMGL+細胞は、主に真皮上層の脈管周囲に存在し、健常皮膚と比較しPsoやADなどの疾患皮膚において有意に顕著な増加を認めた。MGL+細胞が炎症に伴い誘導されるもしくはMGLの発現自体が増加する可能性が考えられる。これらの細胞はCD1c陽性主体の樹状細胞であり、一部のCD1a陽性樹状細胞あるいはマクロファージにも発現すると考えられる。さらに、MGL+細胞数が、ADの病勢及びTh2型免疫応答の指標となる血中TARC/CCL17及び血清IgE値と正の相関が認められた点は注目され、ヒトMGL+細胞のTh2応答との関わりが新たに示唆され、病態解明に迫る有意義な結果が得られた
